Gene therapy iฬถsฬถ ฬถcฬถoฬถmฬถiฬถnฬถgฬถ is here ๐Ÿงฌ
The first genetically modified humans are already walking among us
Building more muscle, losing fat, and de-aging ๐Ÿ’ช
For $25,000, you too can get gene therapy ๐Ÿ’‰
What's the catch?
This is the ultimate gene therapy thread ๐Ÿงต๐Ÿ‘‡
Before we dive in, the usual:
This is not medical advice
I am not affiliated with these gene therapy people
Learning about it just like you guys are
They make lofty claims, we will see if their claims hold true
All I know is this is exciting!
๐๐€๐‚๐Š๐†๐‘๐Ž๐”๐๐ƒ:
Ever since we discovered that DNA is responsible for all our proteins/processes and its a 4 letter CODE, one thing was clear:
We could edit our DNA and influence our biology
This has been the goal for decades. Now it's becoming a reality
This has SCARY implications
Are we now governing our own evolution? Will those who can afford it make the rest of us a sub-superhuman, enslaved species? What nefarious actions will be taken with this technology? Will it turn into a weapon?
sci movie GATTACA explores this:
We have been gene editing viruses, bacteria and other lab animals for decades now
Ex: the drug insulin is made by loading up the gene for insulin into a bacteria
Bacteria then make tons of copies of the drug and is purified
But humans, comes with it risks and concerns..
Gene editing was promising for humans in the 90's
Goal: Genetic disease where someone was missing a key protein
Done through with a Adeno-Associated Virus
1. Load target healthy gene into virus
2. Infect person with ๐Ÿฆ
3. Gene is inserted into person restoring function

If that sounds scary... that's because IT IS
A young man named Jesse Gelsinger received this therapy for his metabolic disorder
His immune system hyper reacted to the virus and he went into shock
Was the first gene therapy death..
This put the breaks on the gene therapy hype
That is... until now. Gene therapy is back with a different technology
Instead of using viruses, new approaches use bacterial plasmids like we discussed with the insulin
Plasmid: extra piece of circular bacterial DNA
1. Target the protein you want
2. Locate genetic sequence that codes for that protein
3. Load sequence onto a plasmid
4. Insert plasmid into bacteria
5. Let the bacteria divide, making millions of copies of the
6. Purify and inject into human
Simple enough?

The gene therapy that is going viral is for Follistatin by a company called "MiniCircle"
This approach can have both cosmetic/aesthetic appeal for some as well as therapeutic implications for others
Let's dive deeper ๐Ÿ‘‡
What is follistatin:
Peptide that blocks myostatin & activin, also decreasing FSH
(statin as a root means to inhibit/block)
1. Follistatin inhibits myostatin
2. Myostatin inhibits muscle growth
3. Inhibit the inhibitor and you get: MORE MUSCLE
๐Ÿ’‰ = you make more follistatin
Animals and humans born with less myostatin are significantly more muscular
(I have one MSTN myostatin gene mutation myself)
What's the goal with this therapy?
Company claims:
- increase muscle mass
- less body fat
- reduction of chronic inflammation
- epigenetic age reversal
In rats, follistatin gene therapy extended lifespan by 34%
We will see if this holds out in humans as they research it
This is appealing for two people:
1. Millionaire biohackers who want to get more jacked and live to 180
2. People born with genetic muscular disorders as well as elderly frail people with sarcopenia
Your favorite biohackers like Dave Asprey & Bryan Johnson already got it
Arenโ€™t we scared of what happened to Jesse Gelsinger
According to MiniCircle, this isnโ€™t a concern:
Plasmids are supposed to be safer and less 'permanent' than virus gene therapy
And the therapy has a kill switch
In front of the gene they put a binding spot for an antibiotic tetracycline
If you take tetracycline, it will turn off the entire plasmid
This makes it reversible
I spoke to someone who received this therapy
They told me:
- became leaner, more muscular and neurologically stronger without training
- No side effects reported yet
- Epigenetic age lowered 7-9 years
- All labs so far have been normal
(As of October 2023 ~250 patients)
Here are the TLDR details:
- one time subcutaneous injection ๐Ÿ’‰
- costs 25k ๐Ÿค‘
- lasts 1.5- 2 years
- leaner, more muscular and "lowers epigenetic age" ๐Ÿงฌ
- Done by Dr. Adeel Khan in ๐Ÿ‡ฒ๐Ÿ‡ฝ๐Ÿ‡ฏ๐Ÿ‡ต๐Ÿ‡ฆ๐Ÿ‡ช
- Clinical trials underway ๐Ÿฅผ
- reversible by taking an antibiotic ๐Ÿ’Š
Is it safe?
The only reported effects so far is a 10 pt increase in LDL level, unclear how significant this is
There is a concern that follistatin can affect fertility given
It lasting 1.5-2 years and being reversible is a +
They are reporting "100% patient satisfaction"
Only time will tell how safe this is
This technology is here and it's about to hit critical mass
They have a goal of making it $5,000 for a treatment, making it accessible to more people
As far as fertility is concerned, they are saying that there are 2 types of follistatin
Follistatin-288 causes infertility
Follistatin-344 (which they are using) does not cause infertility in monkey studies
Before I would get this therapy I would:
- get echocardiogram to check heart size before and after ๐Ÿซ€
- bank my sperm ๐Ÿณ
- full lab panel every 3 months
- DEXA before/after ๐Ÿฉป
- avoid having kids while on the therapy
If this works, this is going to completely revolutionize the fields of medicine, anti aging and biohacking
In the pipeline is gene therapies to increase levels of peptides/hormones like BPC157, GHK-cu and testosterone
According to some sources, athletes are ALREADY receiving this therapy and no agency would be able to catch them...
It's possible that last olympic results were already influenced by this..
Gene therapy is here
Those who can afford take on a giant risk... but also may have a giant edge
Good luck....
